Illumina (ILMN) is set to re-enter the S&P 500 on September 21, reversing a removal that occurred in June 2024. The gene-sequencing firm has rallied 57% from its 2025 low, trading at $206.45. This return to the benchmark index follows a period of significant business recovery and outperformance relative to its mid-cap peers.

The company’s resurgence is anchored in second-quarter results reported on July 30. Revenue reached $1.16 billion, a 9.5% year-over-year increase that exceeded market expectations. Non-GAAP earnings per share of $1.31 also surpassed consensus estimates, marking the fifth consecutive quarter of positive surprises. Management responded by raising full-year guidance, signaling confidence in the durability of recent growth trends.

Clinical segment drives revenue growth

The clinical business now constitutes approximately 65% of sequencing consumables revenue. Excluding China, clinical consumables grew 15% year over year, with the United States and Canada showing stronger gains above 20%. This acceleration is fueled by the placement of the NovaSeq X flagship instrument, with over 95 units installed in the quarter. This pace significantly exceeds the 50-to-60 unit target set 18 months ago, creating a long-term pipeline of recurring revenue.

CEO Jacob Thaysen emphasized that elevated instrument placements indicate customers are building capacity for visible future volume, rather than a one-time transition effect. This strategic shift supports the company’s argument for a durable growth ramp. The clinical segment’s strength offsets broader headwinds, including a 7% decline in research and applied consumables due to cautious U.S. academic funding and continued regulatory pressures in China.

Valuation premium reflects market confidence

Despite the strong operational performance, Illumina trades at a premium to its peers. The stock commands a multiple of nearly 38 times trailing earnings, compared to 23 times for Thermo Fisher Scientific and 22 times for Agilent Technologies. On a forward basis, the multiple is 36 times, with an EV/EBITDA of 22.9 times. This valuation gap suggests the market is pricing in sustained clinical acceleration and margin expansion.

The consensus price target stands at $202.65, nearly identical to the current trading price. This convergence indicates that analysts view the current valuation as fair given the company’s near-monopoly position in sequencing. While the index re-entry will trigger mechanical buying, the stock’s future trajectory depends on maintaining its 22.5% non-GAAP operating margin and sustaining the instrument placement rate.

Cost structure offers structural advantage

CFO Ankur Dhingra highlighted a key structural advantage: Illumina’s instruments do not rely on GPU architectures. As memory and compute costs rise across the hardware sector, this design choice provides a cost edge. This efficiency supports margin stability even as the company scales new product lines. The avoidance of high-cost compute components distinguishes Illumina’s hardware economics from competitors who are increasingly exposed to rising input costs.
